Procedure

1. Label three petri dish containing nutrient agar A, B and C respectively.

2. Transfer 1 cm of Bacillus sp. bacteria into each petri dish and spread evenly
on the agar.
3. Close the petri dishes with lids and secure them with cellophane tape. Invert
the petri dishes.
4. Keep petri dish A in a refrigerator at 5 C, petri dish B in an oven at 37 C and

petri dish C in an oven at 70 C for three days.
5. After 3 days , observe the number of colonies in each petri dish and record
in a table.
