7. Simpan kesemua piring Petri dalam keadaan telangkup di dalam inkubator pada suhu 37°C
selama tiga hari.
Keep all the Petri dishes in inverted condition in an incubator at 37°C for three days.
8. Rekodkan pemerhatian selepas hari ketiga.

Kaedah 1. Tiga piring petri steril disediakan dan dilabelkan sebagai A, B and C.
Method
Three sterile petri dishes were prepared and labelled as A, B and C.

2. 10 cm agar-agar nutrien steril dituangkan ke dalam setiap piring Petri.

10 cm sterile nutrient agar were poured into each Petri dish.

3. Seorang murid dipilih bagi mewakili tiga jenis keadaan tangan, iaitu tangan yang tidak dibasuh,
tangan yang dibasuh dengan air sahaja dan tangan yang dibasuh dengan air dan sabun.
One student were chose to represent three conditions of hands which are unwashed hands, hands washed with
water only and hands washed with water and soap.
4. Tapak tangan kiri yang tidak dibasuh diletakkan pada permukaan agar-agar nutrien steril
di dalam piring Petri A selama 1 minit.
The unwashed left palm was placed onto the surface of sterile nutrient agar in Petri dish A for 1 minute.
5. Tangan dialihkan dan piring Petri A ditutup dengan penutupnya dan direkatkan dengan
serta-merta.
The hand was removed and the Petri dish A was covered with lid and sealed immediately.
6. Langkah 4 dan 5 diulangi dengan meletakkan tapak tangan kiri yang telah dibasuh dengan air
pada permukaan agar nutrien steril di dalam piring Petri B, kemudian tapak tangan kiri yang
telah dibasuh dengan air dan sabun pada permukaan agar nutrien steril di dalam piring Petri C.
Steps 4 and 5 were repeated by placing the left palm that has been washed with water onto the surface of sterile
nutrient agar in Petri dish B, then the left palm that has been washed with water and soap onto the surface of
sterile nutrient agar in Petri dish C.
7. Kesemua piring Petri disimpan dalam keadaan telangkup di dalam inkubator pada suhu
37°C selama tiga hari.
All the Petri dishes were kept in inverted condition in an incubator at 37°C for three days.
8. Pemerhatian direkodkan selepas hari ketiga.
