Bahan Kultur bakteria (Bacillus subtilis), bubur nutrien steril dan kapas steril.
Materials Bacteria culture (Bacillus subtilis), sterile nutrient broth and sterile cotton wool.
Radas Tabung uji steril, silinder penyukat steril dan picagari steril 1 cm³.
Apparatus Sterile test tubes, sterile measuring cylinders and sterile 1 cm³ syringe. K1P4

1. Masukkan 5 cm bubur nutrien steril dan 1 cm kultur bakteria ke dalam tiga tabung uji
steril berlabel A, B dan C seperti yang ditunjukkan dalam rajah.
Add 5 cm of sterile nutrient broth and 1 cm of bacterial culture into three sterile test tubes labelled A, B and C as

shown in the diagram.
2. Sumbat mulut setiap tabung uji dengan kapas steril.
Stuff the opening of each test tube with sterile cotton wool.
3. Letakkan tabung uji A ke dalam almari yang bertutup.
Place test tube A inside a closed cupboard.
4. Letakkan tabung uji B di atas meja di dalam makmal.
Place test tube B on the table in a laboratory.
5. Letakkan tabung uji C di bawah cahaya terang lampu meja.
Place test tube C under the bright light of a table lamp.
6. Perhatikan kekeruhan bubur nutrien selepas dua hari.
Observe the cloudiness of the nutrient broth after two days.
