What is Passive Reference
Dye? (e.g. ROX,Fluorescein dye etc.)
Passive reference dye is used as a reference point to correct the uorescence signal in real-time PCR or to calculate the uctuation
of the uorescence signal. Calibration means modifying to minimize the di erence between the total volume or the equipment
scanner during the reaction. In general, most real-time PCR equipment uses ROX dye as a passive reference dye. This is because
ROX dye does not have any e ect on real-time PCR reaction and emits a uorescence signal that is clearly distinguished from probe
uorescent dye (reporter or quencher). As with some equipment from Bio-Rad, there are some that use uorescein dye and there
are systems that do not require a passive dye such as the Rotor-gene. It is recommended to select the passive reference dye to be
used after considering the advantages and disadvantages of each device or method.
